Applications

G5 Supplement is an optimised serum free supplement which is used in combination with NeuroMix for:

  • Differentiation of ES cells into neuron lineage (neuron and astrocytes)
  • Differentiation of neuronal stem cells into astrocytes and neurons
  • Optimal growth and long-term survival of rat hippocampal neurons (foetal and adult)
  • Survival of neurons from embryonic rat striatum, substantia nigra, septum and cortex, and neonatal rat cerebellum (foetal and adult)

Shelf Life and Storage

G5 Supplement is stable for 1 year when stored at ?-15°C.

Instructions for use

G5 Supplement is a 100 fold concentrate. Dilute G5 Supplement (100x) into the base medium 1:100. The final concentration of G5 Supplement corresponds to 1x. For preparation of 100 ml medium add 1 ml G5 Supplement (100x) into 99 ml of the appropriate base medium.

Cell culture vessels must be coated with poly-D-lysine (0.05 mg/ml). If using N2 Supplement in addition add fibronectin at a final concentration of 5-10 µg/ml directly to the medium.

For cultivation of adult and postnatal neurons

Add G5 Supplement (100x) and NeuroMix (50x) to Neuronal Base Medium AD (add 0.5 mM L-glutamine) to obtain a final concentration of 1x.

For growth of primary and tumor lines of astrocytic phenotype

Add G5 Supplement (100x) to Neuronal Base Medium AD (add 0.5 mM L-glutamine) or DMEM/ Hams F12 (with 0.5 mM L-glutamine) to obtain a final concentration of 1x.
