Medium for cultivation of peripheral blood lymphocytes

QUANTUM PBL is a complete medium stimulating the growth of peripheral blood lymphocytes used for preparation of intact chromosomes. It contains the mitogen phytohaemagglutinin (PHA) as the growth deter­mining factor.
Blood cell karyotyping of lymphocytes is an important tool in modern human cytogenetics to detect chromo­somal aberrations. For cytogenetic analysis peripheral blood lymphocytes are cultivated in specific media and intact chromosomes are prepared. Lymphocytes are mature cells that do not normally undergo subsequent cell divisions. In the presence of a mitogen lympho­cytes are stimulated to enter into mitosis by DNA repli­cation. After 48-72 hours, a mitotic inhibitor is added to the culture to stop mitosis in the metaphase stage. After treatment with hypotonic solution, fixation and  ­observed and evaluated for abnormalities. For short-term ­culture of lymphocytes the use of a mitogen to induce mitosis in non-dividing cells is required. The most commonly used mitogen is phytohaemagglutinin (PHA) which is a red kidney bean extract that transforms lymphocytes. By testing the phytohaemagglutinin in cytogenetic labora­tories the PHA concentration is always optimized for best stimulation of lymphocytes.

QUANTUM PBL is PAA´s complete medium for the selective growth of peripheral blood lymphocytes. It consists of a modified base medium containing high quality phytohaemagglutinin (PHA), pre-tested FBS, L-glutamine and antibiotics.

With QUANTUM PBL a large proportion of lymphocytes are stimulated for growth. Adequate cells are therefore available for further preparation of intact chromo­somes for karyotyping. Consequently, clear karyograms to detect chromosome abnormalities can be drawn.

Extensive quality control tests assure constant high product quality. PAA manufactures cell culture pro­ducts according to current GMP. Further quality control and quality assurances are carried out to the high­est standards as required by the biopharmaceutical industry. Every batch of Quantum PBL is tested by an independent cytogenetic laboratory for its capacity to stimulate the growth of peripheral blood lymphocytes. The resulting karyograms are compared with reference standards.
