Medium for cultivation of amnion and chorionic villi cells    

General

Amniocentesis and Chorionic Villus Sampling (CVS) are important prenatal diagnostic tests. They both involve culturing of isolated foetal cells, followed by chromo­some analysis to detect chromosomal abnormalities.
Amniocentesis is usually performed between 14 and 20 weeks of the pregnancy. Cells from the fluid are examined for signs of chromosomal abnormalities, defective enzymes or deficient growth. More than 75 abnormalities can be detected by analyzing amniotic fluid cells.
Chorionic Villus Sampling (CVS) can be performed ­between 9.5 and 12.5 weeks gestation. This test is done during pregnancy to detect chromosomal abnor­malities and inherited diseases. The most common test employed on cells obtained by CVS is chromosome analysis to determine the karyotype of the foetus.
For growth of amniocytes and chorionic villi cells the use of pre-tested complete media is absolutely essential.

Quantum 3-21 for selective and fast Growth

PAA in cooperation with a number of institutes for human genetics has developed a complete medium for in vitro cultivation of amniocytes and chorionic villi cells. By adding different hormones and novel growth factors, considerable improvements in the growth cha­racteristics of amniocytes and chorionic villi cells have been achieved. Only selected batches of FBS fulfil the requirements of cultivation of amniocytes and chorio­nic villi cells. Batches used for PAA´s QUANTUM 3-21 are determined by extensive tests.
This novel medium recipe enables rapid growth while simultaneously inhibiting other cell types. It is tested regularly by two independent cytogenetic laboratories for its capability to support the growth of primary amniocytes and chorionic villi cells.
With QUANTUM 3-21, clear karyograms for the identi­fication of chromosomal abnormalities can be drawn.

Composition

QUANTUM 3-21 consists of a modified Alpha-MEM supplemented with pre-tested FBS, hormones, growth factors, L-glutamine and gentamicin.
