Stimulation of Lymphocytes

Normally lymphocytes from the peripheral blood are non-dividing cells that have to be stimulated by the most commonly used mitogen phyto­haemag­glutinin (PHA), a lectin extract from red kidney beans (Phaseolus vulgaris). The protein consists of two molecular species, a leucoagglutinin (PHA-L) and an erythroagglutinin (PHA-E). Both species include a ­family of five isolectins. Phytohaemagglutinin has a number of physiological effects and is used in medical research. It not only induces mitosis but also effects the transport of proteins across the cell membrane. PHA can be toxic if used in too high concentrations.

PHA is a naturally occurring product and is subject to batch variation problems. To minimize these variations PAA provides a pre-tested, ready to use solution with a constant and defined concentration of PHA. Every batch of PHA is tested by an independent cytogenetic laboratory for its capacity to stimulate the growth of peripheral blood lymphocytes. The resulting karyo­grams are compared with reference standards. PAA determines the optimal concentration by different quality control tests to offer you a defined ready to use solution because:
