Requirements for Transfection Reagents

Transfection reagents differ considerably in trans­fection efficiency. Most reagents transfer DNA ineffici­ently as cells only take up particles of a specific size. In addition, most reagents cannot sufficiently protect the DNA-complex against endonuclease degradation. New carrier molecules developed by PAA allow the DNA-complexes to be taken up more efficiently, thus increasing the transfection rate.

The Development

Nanofectin consists of two components, a positively charged polymer which is coiled into a porous nano­particle. The specific particle size and structure of Nanofectin allows DNA to be taken up by cells more efficiently. By the principle of surface area enlarge­ment, the DNA binding capacity is increased.  

Uptake of the DNA-nanoparticle Complex

Modern nanotechnology enables the production of  nano­particles of uniform size for an optimized uptake of the DNA-nanoparticle complex.  

Nanoparticles protect DNA

After transfection and release of the DNA-nano­particles into the cell cytoplasm, the structure of Nanofectin protects the bound DNA against endo­nucleolytic degradation. This allows for successful transfection using lower quantities of DNA.  

Nanofectin for stable and transient Expression

DNA bound to Nanofectin can pass the nuclear membrane. Within the nucleus, stable integration into the chromosome can occur. Transient expression in the cytoplasm is also extended with the use of Nanofectin.  

Easy Transfection

Nanofectin is extremely easy to use. The DNA has to be diluted with PAA’s Diluent and incubated with Nanofectin for 30 minutes before the mixture is added to the appropriate cells. The expression is measured after 24-48 hours. Nanofectin can be used in the ­presence of serum so there is no need to rinse cells before incubation with the Nanofectin-DNA comple­xes.
