General
Growth of neuronal cells of the nervous system can be inhibited by some components of Foetal Bovine Serum (FBS). In order to avoid inhibitory effects of FBS neuronal cells must be cultivated under serum free conditions. In the past different serum free supplements have been developed. The N2 Supplement belongs to a small and successful group of multi-purpose serum free substitutes for growth of sensitive cells of the nervous system. It is qualified for cultivation of primary embryonic neurons and optimized for growth of neuronal stem cells.
Applications
- Cultivation of primary embryonic neurons
It is recommended to combine N2 Supplement with the Neuronal Base Medium.
Proliferation and Differentiation
In vitro grown neuronal stem cells provide the opportunity to study basic development together with the possibility of neuronal re-implantation experiments. Serum free cultivation of neuronal stem cells is required for detailed studies. With N2 Supplement, one of the most successful multi-purpose serum free supplements, neuronal stem cells can be grown under defined conditions. N2 Supplement can be used for proliferation of neuronal stem cells. The proliferation rate can be increased by addition of Neuronal Stem Cell Supplement.
For differentiation of neuronal stem cells N2 Supplement must be combined with B27 NeuroMix, a serum free supplement with vitamin A which is necessary for neurogenesis. Depending on the cell types N2 Supplement can be substituted by G5 Supplement. It is recommended to combine N2 Supplement with DMEM or DMEM/Ham’s F-12.
Composition
N2 Supplement contains recombinant insulin, human transferrin, sodium selenite, putrescine and progesterone.
