General
Mesenchymal stem cells, part of the hematopoietic stem cell network, have become a more and more important part of research into human therapeutics. Mesenchymal stem cells, derived from bone marrow, are capable of self-renewal and differentiation into bone-, cartilage-, muscle- and fat cells. Differentiation into distinct cell types can be achieved by directed stimulation using cytokines. In vivo tissue repair using differentiated cells is possible.
Applications
Mesenchymal stem cells can be prepared after separation of mononuclear cells by density gradient centrifugation. Proliferation of mesenchymal stem cells can be increased by using media enriched with specific growth factors from FBS. MesenchymStem Medium developed by PAA provides the optimal concentration of growth factors needed for expansion of mesenchymal stem cells and their progenitors.
For further differentiation into adipocytes, chondrocytes or osteocytes different additives, such as dexamethasone, isobutyl-methylxanthine, ascorbic acid phosphate and indomethacin must be added to MesenchymStem Medium.
Composition
MesenchymStem Medium is a modified DMEM medium with specific trace elements, FBS and L-glutamine. For differentiation of mesenchymal cells into bone-, cartilage-, muscle- and fat cells cytokines must be added.
