Chromosomal Analysis

The karyotyping of chromosomes isolated from lymphocytes, amniocytes and chorionic villi cells is an important tool in cytogenetics. For chromosomal ana­lysis peripheral blood lymphocytes and amniocytes are cultivated in media specifically designed to maximize the yield of intact chromosomes.

Cultivation of Amniocytes and Chorionic Villi Cells or Lymphocytes

Ham's F-10 is one of the most frequently used cul­ture media in cytogenetics. To cultivate amniocytes and chorionic villi cells, important supplements such as foetal bovine serum (FBS) and antibiotics need to be added. However, if you prepare your own media, a considerable amount of time is spent on testing the FBS. Ham's F-10 Ready Mix is a ready to use medium consisting of Ham's F-10 basal medium, FBS (pre-tested on amniocytes and chorionic villi cells), L-glutamine and gentamicin. It is a cost-effective alter­native to other special media used in cytogenetic labo­ratories. This complete medium, when supplemented with phytohaemagglutinin (PHA), is also suitable for the rapid growth of peripheral blood lymphocytes.

FBS pre-tested of Amniocytes and Chorionic Villi Cells

The reliability, safety and quality assurance testing of FBS is performed in two independent cytogenetic labo­ratories. Primary amnion cell growth is screened and the performance is compared to a reference standard.
