General
Hypothermic preservation is a procedure to stabilize tissues and organs at +2°C to +8°C. The goal of cold storage is to induce hypothermia in order to suppress the rate of cell deterioration. An optimal preservation solution contains components to reduce cell-swelling (colloids), buffer to maintain pH balance, antioxidants to scavenge free oxygen radicals and ATP precursors to provide energy upon reperfusion. The cold storage of tissues and organs with optimized hypothermic preservation media is a prerequisite for successful organ transplantation.
For best results, rinsing of biological material prior to hypothermic storage is recommended to remove harmful agents released in the process of extracting.
Applications
For successful tissue and organ transplantation the individual cells must be preserved with the highest viability to maintain the functionality of the whole organ. In contrast to single growing cells only the outermost cells organized in tissues and organs are provided with essential nutrient components, whereas the innermost cells are supplied only by diffusion.
In order to reduce the damaging effects of proteases, nucleases and free radicals CoolStar contains specific inhibitors and components for basic maintenances of the cellular metabolism. Only these specific components allow a high diffusion rate into innermost cell layers assuring optimal supply and preservation.
CoolStar
CoolStar has been formulated to decrease the free radical accumulation in tissues and organs. Free radical accumulation leads to cell death in the case of hypothermic preservation. CoolStar contains inhibitors of proteases and nucleases, and is a further development of the well-known UW-, Celsior- and HTK- solutions. CoolStar has all the advantages of the preservation medium "ViaSpan” combined with further formulations to increase the viability prerequisite for successful organ transplantation experiments.
CoolStar Rinse, the optimized rinsing solution
After the extracting process it is essential to remove potentially harmful substances that could increase the deterioration process of the tissue. Therefore CoolStar should always be used in combination with CoolStar Rinse to permit ideal conditions for long term organ storage. It has been formulated as a flush solution to rinse culture media and native fluids from mammalian cells, tissues and organs.

For optimal results CoolStar should be used in combination with CoolStar Rinse. Both products are available in our cost saving CoolStar Kit.
Composition
CoolStar is a hypothermic medium consisting of disaccharides (sucrose, mannitol, and glucose), lactobionate, dextran, buffers and other reagents which reduce free radicals. CoolStar Rinse is a solution for rinsing of organs and tissues consisting of different disaccharides and buffers.
