General
Various semi-solid culture systems have been used for detection and differentiation of hematopoietic stem cells. Both methylcellulose-based media and collagen-based media are available.
Methylcellulose-based cultures cannot be fixed for subsequent cytochemical or immuno-cytochemical staining. Therefore collagen gels are used for this application. Collagen-based media are an attractive alternative to methylcellulose-based semi-solid cultures. Collagen, a gelling agent, increases the viscosity of the medium allowing optimal colony formation of different progenitor cells for simplified separation and identification. Collagen can be fixed for cytochemical or immuno-cytochemical staining and the resulting slides can be stored long-term.
Composition
The CollagenStem Medium consists of a modified IMDM medium containing BSA, 2-mercaptoethanol and L-glutamine. This medium has to be supplemented with the CollagenStem Solution.
Furthermore FBS (10–30%) and cytokines can be added.
