General

Cells of the central nervous system need specific grow­th factors for long-term viability. Additionally more vitamins and fatty acids can enhance the long-term cultivation. The recently developed B27 NeuroMix is a multi-purpose serum free supplement for long-term viability and growth of hippocampal and other neurons of the Central Nervous (CNS) and Peripheral Nervous System (PNS). B27 NeuroMix contains also specific growth factors for differentiation of neuronal stem cells and differentiation of murine embryonic stem cells into neuronal lineages. It is based on the neuron-specific B27 Supplement developed by Brewer but with an improved formulation.

Applications

For detailed studies of neurons defined nutrient media for growth are necessary. Basal media supplemented with foetal bovine serum contain factors which can inhibit growth of neuronal cells. Therefore specific serum substitutes were developed for optimal survival and growth of embryonic and adult neurons to elimi­nate neurotoxic side effects.

When added to Neuronal Base Medium, B27 NeuroMix supports the growth of neuronal cells without the need of specific feeder cells.

Differentiation of neuronal Stem Cells

For differentiation of neuronal stem cells the supple­ment contains Vitamin A. B27 NeuroMix can also be used together with G5 Supplement to improve growth and differentiation of neuronal stem cells into glial cells and neurons. For differentiation of murine embryonic stem cells into neuronal lineages B27 NeuroMix and G5 Supplement provide essential growth factors and hormones.

Composition

The supplement is a chemically defined 50x concentra­te and contains vitamins, hormones and other growth factors including insulin, transferrin, catalase, superoxi­de dismutase, antioxidants and fatty acids.
