Treatment with MycoKill AB includes three or more cycles (passages). It is recommended to check the culture every week for presence of mycoplasma (e.g. ELISA, fluorescence staining or PCR). Normally mycoplasma will be removed after 2-3 weeks.
Shelf Life and Storage
MycoKill AB has a shelf life of 2 years when stored at ≤-15°C.
Working Concentration
- MycoKill AB: 50 x stock solution in PBS.
- Working concentration: 1x (final 10µg/ml in medium)
- Dilute the stock solution 1:50 (Step 1)
- Example: add 10 ml MycoKill AB to 500 ml base medium
- If required increase MycoKill AB as follows in different subsequent steps:
- Step 2: dilute 1:40
- Step 3: dilute 1:30
- Step 4: dilute 1:25
Instructions for use
- Step 1 (with 1:50 dilution)
- Remove culture medium from culture vessels, wash the cells with PBS and detach cells with Trypsin or Accutase
- Count the cells and plate in fresh medium (with MycoKill AB)
- Cultivate the cells for 2-3 days in the usual way
- Remove culture medium from culture vessels, wash the cells with PBS and detach cells with Trypsin or Accutase
- Count the cells and plate in fresh medium (with MycoKill AB)
- Cultivate for 2-3 days
- Repeat the passaging cycle at least twice
- Check for mycoplasma contamination (Staining method, cultivation or ELISA).
- If mycoplasma positive, check methods (positive and negative controls and evaluate)
- If required increase MycoKill AB described above (follow Step 2, Step 3 etc.) and repeat treatment with increased MycoKill AB concentrations.
